The anti-SD sequence (the 3’end sequence of 16S ribosomal RNA) will pair with the SD sequence (5’ untranslated region) for the precise translation. This binding interaction will form a “correct” translational initiation complex in e coli system. In the paper, we have found there are many questionable 16S ribosomal gene annotations based on ambiguous sequences. Such wrong annotation have already blurred the accuracy of present database.
